I am planning a cycle trip that will start in Kathmandu and head East towards Darjeeling.
Does anyone here have any info on the difficulties of this, such as hills and availability of lodges/guest houses?

You have three options out of Kathmandu to the plains (Terai): The mountain road via Daman to Hetauda is super scenic, though a big climb. You could also follow the main highway to Bharatpur and Chitwan National Park. The shortest is the relatively new road east via Dhulikhel; I've not done this one.

Janakpur is an interesting detour on the plains for its culture.

You should be fine with finding places to stay, but best to get an early start each day in case distance to next accommodation is far.

From Siliguri, you can take the road to Darjeeling that follows the toy train route. This has gentle grades, unlike the other options.

Have a look at guidebooks or Internet. You can also check www.crazyguyonabike.com for travelogues and ideas.

Hello Bill, we met a couple of years ago on the road somewhere in Thailand.

I had already searched on Crazyguy and other cycling forums but found nothing. I am on this trip now and have just done the bit from Dhulikhel to the Terai. Now enjoying a rest day in Bardibas.

It is really a very nice route to ride. Very hilly but nice. I have had no problems finding either guest houses or food.
Along the way I have stayed overnight at Nepaltok, Kurkhot and Sindhuli Madi. All of them within a reasonable daily ride. For the remainder of this trip in Nepal I will be on The East-West highway and expect no problems finding a place to stay etc. I hope and expect that in the future there will be more cyclists here. I have not seen any others and have attracted quite a lot of attention from the locals. All sweet and helpful people.

I rode the old highway to the Terai about 20 years ago. TheTerai was a real eye opener as I hadn't realised Nepal had flat bits.

Much as I loved India and had been many times, it was a real shock after easy going Nepal

Enjoy.
