I am intending to hire a car in Pisa and, a week later, leave it in Florence because I'm staying there for a few days. Would it be easiest to return it to Florence airport and get the bus into the city or take it into one of the car hire places in central Florence?

I would strongly recommend returning the car at Florence airport and taking the bus into town. Driving in Italian cities is no fun while the airport is outside the city center. We rented a car in Venice, drove around Tuscany for a week then returned the car in Florence airport. We found the airport bus very fast, easy and reasonable (€4 in 2004).

Agree totally with sonichaggis. We found driving in Florence a nightmare. N.B. The airport is at Peretola but is often referred to on road signs by the term "Aeroporto A. Vespucci".

What are you doing in Florence? If you are not traveling that much, I would not rent the car at all. Parking can be a nightmare in Florence.
