What is the best way to get from the airport to the Grand O hotel Omonia Square. Taxi, subway, bus, etc.

The subway (metro). Airport to Syntagma sq. then change to Omonia (red line)

from airport to town, you take it in the direction of Monasteraki or Egaleo [the new terminal station just opened up and now the new terminal instead of Monasteraki], get off at Syndaga as #1 states for Omonia [in the direction of Agios Andonios]
