The road to Shatili branches off the Tbilisi - Kazbegi highway in Zhinvali. I wouldn't call the route direct, but you don't need to go back to Tbilisi.

Kazbegi to Zhinvali 2 hours. Zhinvali to Shatili - I would guess something around 5 hours. Generally I would reserve a whole day for this route, the road on the section Barisakho - Shatili is very rough.
