What do you mean "best way?" Do you mean type of transport - taxi or bus? Or something else? Have you ruled out flying direct from Delhi? @vistet may have advice

Hey I am looking for adventures route and same time I am looking to go by Taxi.
I really don't want to fly but I might take flight when flying out...
The thing with going from Srinagar, is it will take you a long time to get there. Once there, it's still a 2 day trip. Manali is easier to reach from the south, and there are many more tourist on the Manali route.
The Srinagar route is better for acclimation and has an interesting site on the way. Manali has the greenery if you want to see what leaves look like..

Definitely Srinagar for better acclimatization as altitude rises gradually you can also visit drass and kargil war memorial on the way and lamayaru monastry.

Delhi to Manali is a overnight bus. Delhi to Srinagar starts with a overnight train ( a lot comfier ) to Jammu , and taking one of the cabs waiting outside the station to Srinagar - you´ll arrive in the afternoon . Nice ride. There is a new tunnel opening in July that will make it a bit faster.
Once there, it's still a 2 day trip.
Standard run with the shared cabs that you find at the Tourist Reception Center is one long day to Leh . You lose some great views enroute coming in to the Indus Valley in the evening/night. Nearly all of the acclimatization advantage also comes from stopping overnight in Kargil @ 2693 : nights well above 2000 is what really matters . On this route its mostly a comfort issue though . You may get sick after arriving in Leh , but not dramatically so.
Manali-Leh runs a lot higher - the tent camps at Sarchu & Pang are higher than the passes on Srinagar-Leh , and max altitude is the same as Everest Base Camp , 5300+ meters. You do not want to try how susceptible you are to altitude sickness by doing the cannonball Manali-Leh run - think up to twenty hours when everything works out . Or two days : over the years I´ve made two unplanned overnight stops in Pang , more than a thousand meters higher than Leh , and I was helping/dragging someone out of the vehicle both times. Stop instead overnight in Sissu , Keylong or Jispa , all around 3100 meters . More pointers here : http://korta.nu/sleepless
