I am going to Eastern Maryland for the first time, as Duo, and only for 2 full days & 2 Nights, and am looking for any highly recommended places to see, visit, drive through, eat, beach, historical sights, etc., that are special and/or unique to this region.

We will be driving from Annapolis, over the Bridge to Queenstown, and then South and then East after that. We want to explore as much as possible. We are Not going through Delaware.

I'd like to see as much of the original beauty, towns and structures as possible for the driving part of the trip. The first night we stay in Salisbury and the second night we stay close to Assateague. We will definitely end the trip at Assateague, and then drive to Virginia Beach and up to Richmond, VA.

I thank you in advance for any advice on the above that you can suggest and that you think are the best parts to visit/see on the Eastern Maryland Peninsula.

This is my old stomping ground. I used to live in Salisbury, and also a little town called Preston.

Your best itinerary for the first day is from Easton, detour to St. Michael's, get out of the car, and explore that pretty town, including a stop in the Chesapeake Maritime Museum. Then you can drive south onto the neck to Bellevue, and take the ferry to Oxford. And get out of the car and explore that pretty town. Then drive back out to route 50.

I would be tempted to stay in Cambridge the first night, instead of Salisbury. Cambridge is a bit more atmospheric, and the waterfront is prettier. Salisbury is bigger and more commercial, and only 45 minutes from Cambridge.

Assateauge SP and NP are wonderful. You will love that. On the way to Virginia, the 2 best towns are Berlin and Princess Anne. Though Princess Anne requires a few miles detour north on rt. 13. Or you could drive out to Crisfield on the Chesapeake Bay. But if you go to Crisfield, you have to eat crabs. They won't allow you to leave the town otherwise.
