I can't give you recent information, but i think the trip will last 10 hours or so, there will be either nightbusses or early morning busses. Only ordinary busses, no upmarket bus companies.

Your quickest option and a route that I took earlier this year would be to travel via Moquegua which has a journey time of around 3.5 to 4 hours by combi (35 soles). They depart 100 metres from in front of Puno's Terminal Terrestre on the corner of Av. Simon Bolivar and Jr. Echenique, the latter leads up from Puno's terminal. This is a relativley new asphalted route direct to Moquegua and heads straight inland from Puno and does'nt take you the long way round via the shores of Lake Titicaca via Desaguadero. Once in Moquegua, Transportes Moquegua depart hourly to Tacna, 2 hours, 10 soles, plus there are combi's departing every thirty minutes or so as well as other bus companies. Moquegua is a pleasant town and if you're not in a rush then is worth spending a night.
