I want to travel overland by bus from Lima, Peru to Tucuman, Argentina through Chile or Bolivia, and need advice on the best travel route. The trip should span 6 weeks, so I can see some things, and not be exhausted by long bus rides. I will consider train segments, if they are popular, efficient and affordable. I will be staying at hostels along the way, and would appreciated advice on these also.

Well, that's not just a bus route, but a vacation in which you presumably want to see stuff. Do some research on the stuff you want to see. The most obvious route is Lima - Cuzco - La Paz - Tupiza - Salta - Tucuman, with plenty of stops along the way, and detours. 6 weeks is a decent amount of time for the route with stops, not so good for detours. Look up the things you'll be going past on that route and see what you like.
