I will be in Ecuador next August. Starting from Quito I plan to reach Cuenca via Ambato, Riobamba, Alausi, but I would like to consider a different itinerary on my way back to Quito. The option Cuenca-Babahoyo-Guaranda-Latacunca by bus is possible in one day? Or do I have to move from Cuenca to Guayaquil?

The trip is certainly a very long one, maybe if you have the right connections, you can make the trip in one day, but it won't be fun.
Maybe a little stop in Guaranda would be an idea, it's a great little place, I'm sure you'll have a lot of fun.
Imposible..... would take you about 20hours... From cuenca you´d have to go first north to Latacunga 8 hours, then go back south to ambato 1 hour, then take ambato to guaranda 2 and half hours, then to Babahoyo 2 and half hours, then to Guayaquil three hours... may be more if the road is in bad condition....
This not including stops and all of that... concider another route... don´t go to Guaranda there is nothing there to enjoy, babahoyo is nice for nature and subtropic and rivers though!
Concider rescheduelling or three days to do this.. if not take the easiest way to Guayaquil Cuenca Troncal Guayaquil, round 5 hours!
