Visiting Manaus at end of February.. What's the best transportation from airport to downtown, also would like a boat trip to see the confluence of the the two rivers. Only have 3 days there, so the boat trip would need to be only a day or half day trip. Anything else there of any great interest?

Just take any airport taxi. I wouldn't recommend trying a bus.
Manaus does a rousing business in one-day tours. The Meeting of the Waters only takes a few minutes to get to (that is, if they go straight there) and a few minutes to observe, so you should sign up with a tour that will take you to do more (see TripAdvisor for recommendations)--it'll be pretty touristy and not like the "real" jungle, but should be fairly interesting. If you can afford it make sure you take a private or small-group tour, not one with forty other people--I see those sometimes. Otherwise, you can take a tour of the opera house or at least see it from the outside. There are a few parks, but I don't think it would really be worth it unless you especially love manatees (in tanks). Lots of good fresh fish to see at the market if you like that kind of thing. Try to get fish for lunch or dinner as well, or if you don't like fish, get beef/chicken/pork kebabs at a sidewalk stand. Drink fresh juice and eat ice cream.
