(re: the race using a different route from Flores -> Lanquin) I am very interested to hear if that route is now paved/smooth/available for public transport. Last spring, I wanted to go that other route you suggest, but was told from several sources that it was not possible because of poor road conditions, construction, and lack of buses traversing that route. Has that changed? The sources I received that from were from our guesthouse in Antigua and Lanquin, as well as some bus drivers and locals.
Also, from Flores/Santa Elena to Coban, there are a couple direct buses available - at least there was in the Spring. That way, the op could avoid having to switch minibuses constantly.

is there an option other than Lanquin for an overnight to break up the trip from Flores to San Pedro de Lago Atitlan?
Another route you can go from Flores -> Lake Atitlan would be to go via Rio Dulce & Guate City. As #7 says, there are several hotels nearby you can stay at. Finca Tatin is really pretty and quiet - but you'd need to add a couple days to the itinerary since it is a decent boat ride away. There's another place closer (forgot the name and my internet is slow here in Laos! - but it's on my travel blog) to Rio Dulce town with a similar atmosphere.
