The best spot for wildlife is the Sirena ranger station in Corcovado Park. From Drake Bay you can get there by boat. I don't think you have to book the day trip in advance. But staying overnight at Sirena would be even better (best times to see wildlife are dusk and dawn). That is expensive and if you would like to do that you should probably book it in advance. But your hotel will be able to help out with any tour you might want to take. You can take many other tours and they don't have to be booked in advance: Rio Claro, snorkeling, mangrove tour, night tour...

In Manzanillo just go for a walk and you will see lots of wildlife. I assume you will even see lots of wildlife on the hotel property. Walk to Punta Uva and see more wildlife. Or rent a bike and explore more beaches. There is also the Gandoca wildlife refuge in Manzanillo. You can go for a short hike on your own but if you want to do a long hike there it is best to go with a guide. I would also highly recommend staying in Cahuita for a few nights or at least doing a day trip to the National Park in Cahuita. You can take the bus to Cahuita and walk to the park entrance. You can explore on your own or hire a guide at the entrance. The park has great beaches, a rainforest trail and lots of wildlife.
