The offices might not be open on Good Friday, but if any tours have been per-arranged, they will go ahead with the tour on GF.
The zoo is about 30 km east of Belmopan. You could easily get a Regular bus heading to Belize and ask to be dropped at the zoo. Two hours should be enough at the zoo. Return to the highway and wait for a bus heading back to Belmopan.
