Hello
I'm hoping to spend 2 and a half to 3 weeks in the country in November and would like to do a bit of everything including safari, jo'burg, winelands, cape town beach etc etc, much research needed but any starting suggestions appreciated.

Also is anyone familiar with the weather at that time of year in the different areas?

Safari - the Kruger Park is wonderful in November when the young have just been born and the weather is not yet too unbearably hot.
Johannesburg - Great at any time of the year with plenty to do and see. I have posted a list of things to do in and around Johannesburg so often recently that all you have to do is use the search facility.
Cape Town, Winelands, Beaches - all great in November.
No need to make suggestions, just keep reading this forum.
Oh, you left out the Drakensberg, and the Transkei, and the Kalahari.

Two and a half weeks is not much time to do as much as you list, so you have a choice. Touch ground only on the bumps. You will get a glimpse of a lot of of different things but not have enough time to really enjoy any of them. This will be fine if you plan to go back again soon but if not I recommend narrowing it down to three things and that way you get time to really get a feeling for the country. We spent three weeks in SA last Nov and ended up dropping plans for some things. Not a problem as we're going back next year but my wife who had never been there before says she wouldn't want to do it any other way. We made sure we stayed at least a couple of days in each place and took more time at places like Kruger.
