1Makarska is on the sea level and the top of Biokovo is on 1762 meters above sea level. The route, paved all the way is a bit less than 30 km long and the average gradient is between 10 and 15% and a bit more (up to 20%) before the top of the mountain.
According to the experience of most of the cyclists that have performed the ascent, the route is moderately hard, but every well trained cyclist can do it within the time frame of let's say 3 to 3 1/2 hours... This implies several stops on the route for the rest and to enjoy the scenery.
Here on this short video, recorded by two German cyclists, you can see the average appearance of the route: http://www.youtube.com/watch?v=UJF8SnW6CL0 .
Please, keep in mind that you will have to pay the entrance ticket, but as a cyclist you pay just 50% from the original price. So you will pay 20 HRK for a ticket (roughly 2.7 EUR or 2.3 GBP - http://www.biokovo.com/en/71/price-list ).
This route, from Makarska to the top of Biokovo called Sveti Jure (St. George) is pretty popular among the cyclists, so you can expect that you will not be alone.

2My friend went from Makarska to Biokovo with bike last summer. Cycling route from Makarska to the top of Biokovo is about 30 km long, it took him about 3,5 hours if I remember correctly. If you go during the summer period it is very important to start your route early in the morning, about 6, 7 a.m. so you could finish till 10 a.m. because of very high temperatures.
Surface is mostly paved and not too demanding for a ride. Don't forget to take enaugh water with you because there is no water source on most of the routes. You should also bring windbraker because of the strong wind on the top of the Biokovo.
