I'm arriving into Peru from Puno. I would like to visit Arequipa, Colca Canyon, Cusco, MP and a few other places towards Lima. I've been advised to perhaps get on a 4M Express tour that passes through Puno towards the colca canyon but originates in Arequipa and returns there.I'll be in Peru in May and would like to spend a few days in Arequipa. Does anybody have any advice as to what would be best, considering my onward journey too?

1Best to make all your tour arrangements locally. You can take a bus Puno - Arequipa and then figure out your Colca trip and then travel to Cusco and figure out MP/Valley Sagrado. There will probably also be MP tour companies in Arequipa that could offer some packages as well, and even possibly from Puno.

3You might also look at flights between Arequipa & Cusco, Cusco & Lima, and Arequipa & Lima. When I was in Peru last year in March/April, the flights were only a $20-30 more than the buses on those long routes, and took just a couple hours instead of dozens of hours. The time I saved made it totally worth it, since I was only able to be in Peru for a few weeks.

5Maybe you could visit Cusco & MP first as flights to Lima are cheaper from Arequipa and bus trips are shorter form Arequipa to Lima than from Cusco.
If you like tourist buses you could take one of the several tourist buses from Puno to Cusco, has a couple of interesting stops en route like the remains of Wiracocha's temple in Raqchi and the church in Andahuaylillas with very interesting paintings which remain me Hieronymus Bosch. IMHO this route is more interesting than the one between Puno & Chivay in Colca Canyon.
This is one of the companies to have an idea of the route: http://www.cuscopunobus.com/route-of-the-sun-puno-cusco.html, never took it as I'm not so into this kind of bus travel but I have been to the sites several times and found these interesting.
Don't miss Sillustani when in Puno, interesting funerary towers between Puno & Juliaca.
