I think the most important part of the country, both Historically and Culturally, is the Savannah, Charleston, Colonial Williamsburg, and Wash DC areas. Charleston and Wash DC are both 4 nights easy, DC more just for the Museums. The BRP is lovely, the highlight is from Bryson City on the south end of Smokey National Park, to Asheville via Mount Pisbah area, great hiking there.
Asheville is a very important music and arts area, with the Biltmore Estate a must see.
You can alsodeviate from the main Interstates and drive from this area along the Appalachian Trail areas, this back country and down home traditional Americana.
In regards to the Kentucky Derby, there is also great racing and scenery in Lexington, and the Keenland Track, also Lexington is a more quaint small city to enjoy a few nights. In between Lexington and Louisville, is some excellent areas ranging from the Whiskey Trail Bardstown, wine country and the Shaker Village area, as well Danville, Harrodsburg and great State Parks like Cumberland Falls, Natural Bridge State Park and a whole lot more.
I had trip reports for all these areas but the recent LP TT melt down has them off limits at present.
