Suggestions between Strasbourg and Frankfurt

I did a quick search on this forum and most threads discussing these two cities seem to be about how to get between the two cities without much talk about anything in between.
Any suggestions? Is the Strasbourg/Frankfurt split 3day/2day good? Are there any other recommendations for stops in between that might be better worth my time?
PS. I will have a 12h lay over in Frankfurt when I initially arrive in Europe where I plan to spend around the city, so I will already have seen a little bit.

3Heidelberg is on the way but quite touristic, Stuttgart could be a good option.

6Thanks for the replies, I'll definitely check out some of the suggestions. As for what I'm looking for, I'm not really looking for crazy night life for traditional museums. I'd like to wander around and enjoy the city "feel" (architecture, people etc), enjoy some good eats, and maybe something a little experiential (I like the river cruise, walking tours, brewery tours etc).

7Frankfurt gets a bad name as a tourist destination mainly, I suspect, from people who have only passed through the airport.
In fact its a vibrant modern city which spends more on art and culture per head than any other German city. It also has, according to Lonley Planet, the best nightlife after Berlin of any German city. Add to that a stunning skyline and a good riverside ambience then Frankfurt has more than enough to fill a couple of anyone's days.
