The forum's resident expert on Belize transportation will probably respond to you, but the short answer is yes, it's easily possible.Even if you take local buses, rather than express buses, tourist buses or shuttles, you can go from San Ignacio to Chetumal in eight hours or less, including connection and border crossing times.

Regular Belizean buses to BC bus terminal depart from SI every 30 minutes. The trip is a bit over 2 hours and costs BZ$7.There is also the Valencia bus (one or two per day) that travels from Benque to Corozal, but I think it departs SI in the afternoon.
In the BC bus terminal, change to a bus heading north -- either Corozal or Chetumal will be displayed in the front window of the bus. Buses depart approximately every 30 minutes, the trip to Chetumal is about 4 hours, and the fare is about BZ$12 or $13 to Chetumal. If there is no bus to Chetumal, just get the next one to Corozal and change in Corozal.

The Belizean buses stop crossing the border and going to Chetumal sometime around 6 or 7 pm. You can still use buses to get to the border, walk over, and then use Mexican buses or taxis to get to Chetumal.
