2or Karlstejn Castle...
Both destinations easily accesible by public transport. Kutna Hora - direct bus from Prague Florenc Autobus station. Travel time approx. 1 1/2 hour. One-way ticket approx. 50 Kc's
Karlstejn - direct train from Hlavni Nadrazi (Main train station). Travel time approx. 1 hour, one-way ticket approx. 50 kc's
Personally I prefer Kutna Hora.

3Kutna Hora for a cute town with an amazing castle. If you get up early, you can get a train/bus and visit the Cesky Raj (Bohemian Paradise) area which is forests, huge boulders and 'rock cities' and some old castles, very pretty. Or a daytrip to Terezin which can be combined with a cute town nearby - forgot which.
Use the excellent www.vlak-bus.cz to sort out all train and bus connections, English version available.

4Why not escape the tourist throngs and head for Krivoklat, a fairytale castle perched high up on a rocky promontory above the Berounka River around 50Km west of Prague. The forests and hills around are ideal for walking and the area doesn't get the hordes of tour bus zombies like Karlstejn does. This is the Czech Rep. at its best. Again www.vlak.cz will give you all the information you need about getting there and away. www.krivoklat.cz is the castle's website (in English).

5Agreed - Krivoklat is nice, you can get to it with one change onto a bus-like train, see the castle and do a walk through the forests (there are marked routes and you can take a photo of the hiking map with your digital camera for consulting along the way) ending up at Karlstein castle so that at least you see that one on the outside too. Then hop on a train back to Prague at Karlstein station. Did that last winter, very pretty in the snow.

6It all depends on what you're interested in, but I would recommend Karlovy Vary if you like architecture, Kutna Hora if you want to see the Ossuary and gothic church, and Pilsen or Ceske Budjeovice if you like beer. This site has info on transport to and from Prague, as well as things to do and see.
